Researchers from Italy and Canada reveal that papillary thyroid cancer subtype has longer telomeres than other variants

The most important representative of endocrine malignancies is papillary thyroid cancer. Its special subtype possesses features that are similar to non-malignant lesions, so it is categorized as nonivasive follicular thyroid neoplasm with papillary-like nuclear features (NIFTP). NIFTP genetic profile is still controversial, so researchers from the University of Cagliari, University of Sassari and University of Manitoba jointly designed a study to investigate whether a specific 3D architecture of chromosome ends known as telomeres can be used to potentially distinguish NIFTP from other variants.

Study is based on the fact that in contrast to normal cells, cancer cells have altered 3D telomere assembly. Results of present study demonstrated that 3D telomere architecture of classic and follicular variants differed from NIFTP, and that NIFTP was more similar to a benign thyroid tumor. Namely, NIFTP has longer telomeres than classic and follicular types, which corresponds more to a benign lesion.

Based on these findings, researchers concluded that 3D telomere organization is altered differently in thyroid cancer variants, therefore this feature could have a diagnostic value and might aid the clinical management of NIFTP, which still has indefinite characterization and a difficult to predict outcome.

Il rappresentante più importante delle neoplasie endocrine è il cancro papillare della tiroide. Il suo sottotipo speciale possiede caratteristiche simili alle lesioni non maligne, quindi è classificato come neoplasia tiroidea follicolare non ivasiva con caratteristiche nucleari simili a papillare (NIFTP). Il profilo genetico del NIFTP è ancora controverso, quindi i ricercatori dell’Università di Cagliari, dell’Università di Sassari e dell’Università di Manitoba hanno progettato insieme uno studio per indagare se una specifica architettura 3D delle estremità del cromosoma nota come telomeri possa essere utilizzata per distinguere potenzialmente il NIFTP da altre varianti.

Lo studio si basa sul fatto che, contrariamente alle cellule normali, le cellule cancerose hanno alterato l’assemblaggio dei telomeri 3D. I risultati del presente studio hanno dimostrato che l’architettura telomerica 3D delle varianti classica e follicolare differiva dal NIFTP e che il NIFTP era più simile a un tumore tiroideo benigno. Vale a dire, NIFTP ha telomeri più lunghi rispetto ai tipi classici e follicolari, il che corrisponde di più a una lesione benigna.

Sulla base di questi risultati, i ricercatori hanno concluso che l’organizzazione dei telomeri 3D è alterata in modo diverso nelle varianti del cancro della tiroide, quindi questa caratteristica potrebbe avere un valore diagnostico e potrebbe aiutare la gestione clinica del NIFTP, che ha ancora una caratterizzazione indefinita e un esito difficile da prevedere.
